Yeah lets have a thread about them.
1. If you took say an anti-euphoric drug or something legal but taken in large doses very painful/unpleasant and triggers negative chemicals in your brain, wouldn't the comedown be euphoric? Like think of it as an amphetamine comedown in reverse. I don't know if that makes any sense.
2. If you drank denatured alcohol for long enough, over time wouldn't you build a tolerance to methanol and therefore have an extremely cheap source of strong spirit.
yeah, anyone else have questions? I hope what I wrote made some sense...

Are you, by any chance, high? :p I'll give your questions a go, but I'm not claiming in any way that I'm right, more likely I'm completely off. I'll leave it to Mr Pedo & other members to point out my mistakes. :)

I think it unlikely, taking the example of serotonin as your neurotransmitter, your brain is more likely to try to regulate a constant quantity of serotonin in presynaptic knobs, rather than working on constantly releasing it at an average rate as it is constantly being produced. If it is somehow blocked from being released, as this theoretical drug could do, it wouldn't necessarily make any more serotonin, or particular neurotransmitter. It would therefore not result in a heightened quantity of neurotransmitter being released once the drug had been broken down, sadly.

Sadly not. I don't think your liver will be able to find another way of metabolising methanol that doesn't result in such toxic byproducts. As it stands, methanol is metabolised to formic acid via formaldehyde, formic acid being a fairly nasty chemical to have sloshing round the system.

nah its cool :) This thread didn't really make sense in the first place. But yeah, I just read that I think that some ancient roman of significance or something had small doses of some sort of poison over time and eventually built up a tolerance so that if someone tried to poison him it wouldn't work. As for the other question, I assumed the human brain tried to obtain some sort of balance, and if a drug ruined the balance it'd just try and even things out.
But yeah, if anyone has any ridiculous drug theory questions, heres the thread to do it.
Oh, and as for being high, I'm in thailand at the moment and not much passes for being a 'pharmacist'. I walked into one pharmacy on phi phi island and the lady at the counter had a marijuana leaf t-shirt on. It's kinda funny. :)

1) being relieved from pain is a euphoric experience yes, but is generally overclouded by memories of past experiences.
2) methanol will poison you before you develop resistance, i.e.: you die, blind.

AFAIK, the body's not good at upregulating stuff because of artificial downregulation, but there's probably some way to do it. For instance, I could be wrong, but I thought opiate tolerances are a result of either desensitization of receptors or downregulation of endogenous opioid agonist. After some time off the dope, things return to normal, so the body changes one of these two things in response to something. The questions is, what, and how does one acquire it.
Alternatively, downregulation in response to artificial upregulation is the more common biological response. So, artificially upregulate something nasty , say...kappa opiate activity? But where to get a kappa agonist? Salvinorin A.

probably not, Salvia acts on the kappa opioid receptors, which cause generally dysphoric feelings along with the bizarre hallucinations.... i don't feel all that euphoric coming down from it just a feeling of "what the fuck?"....

you would probably just go blind gradually before you had any tolerance....

It wasn't just Roman times. In medieval times people who were more paranoid [even if they had good reason] would take small amounts of various poisons to build up a tolerance. That way if anyone dosed them with a normal dose, they would only get sick and not die - or so the theory went...
