The first thing you'll want to do is jack your methylation through the roof. Since methylation is handled by s-adenosylmethionine, you'll... probably want a lot of methionine.

'n since it produces homocystine, 'n homocystine tends to regenerate to methionine if the reagents are avaliable, one will probably want to maximize regeneration, so one's methionine levels keep growing, and growing, and...

...one of the regeneration pathways of methionine is through glycine betaine, which the body manufactures from choline... so, jack your choline.

The other two means of regeneration are through folic acid 'n b12, respectively.

So, once one has been adding a gram or two of methionine to the system, and maximizing recycling efficiency, for a day or two - or a week or two - one could probably just drop half a gram to a gram of tryptophan and watch the vast majority of it get instantly alkylated. Of course, few people have put this into deliberate, controlled practice... so it's sort of experimental ground.
