When I have a lot of E I generally like to pop some and snort some throughout the night. In your case I'd like pop one maybe then snort one, then snort the other half later maybe? I don't know if re-dosing MDMA makes a difference though.

If you chew them up effects will come on faster and be more intense, but it won't last as long. Railing them will make it come on MUCH faster, MUCH more intense, and MUCH shorter.

Take them intranasally, don't even think about popping them.

Two and a half pills is a shitload to rail.. Especially since this is someone who has to ask what's most effective. I'm guessing they haven't railed pills before.

No way, snorting E is a complete waste... People who snort pills only serve to give drugs and drug users a bad name. Seriously, if you can't wait for a half an hour to get effects, then you need to reevaluate a lot of stuff in your life. If you want it to hit you faster and much MUCH stronger... plug it. Completely crush the pill into powder, mix it thoroughly in water, and use an oral syringe to inject it a few inches up. There are so many blood vessels and shit there it absorbs much faster and bypasses the first pass liver filtering. I've heard for some stimulants it can increase the effects up to 6 times. So, I'd go with much less then what you'd normally take. I know even for drugs like Oxycodone which has the same bioavailability rectal as orally, it's still stronger to plug. That doesn't make sense to some, but the peak plasma levels are different, etc. It doesn't last as long usually, a couple hours shorter at most, but with how much less you'll need to get high... it's well worth it.
If you don't want to plug it, then just crush the pill into powder and parachute it or just swallow it with a strong flavored liquid like coca-cola (caffeine free of course since mixing stimulants is a bad idea and usually rolls have multiple chemicals in them anyway...) Snorting doesn't feel anywhere near the same, you'll never get that wonderful good euphoric empathy feeling the same way as if you ate it. Besides, it's better to eat them and wait, and you don't feel anything... and then at about a half an hour, ZANG, and suddenly it hits you and you're rolling nicely. That's the way to go. Who wants to destroy their nose, possibly their sense of smell, get an awful tasting drip, possibly have a clogged nose, and maybe even get less out of their pill? I've always felt less potent effects from insufflated MDMA then oral consumed however there are unfortunately no studies I've found with verified bioavailabilities of one or the other.
