I usually just talk to the girl for a while and get a read from there. Body language can show a lot about girls, even if they don't admit to it. Every girl's body language is different though, so it's just trial and error.

Once you can start reading a girl's body language, you can tell what she's thinking and you can pick up on if she wants to hook up with you or not. I really hate Public Displays of Affection when I'm at parties and such, because it gives the girl too much power. I like to change locations until I feel we're in the right environment. If she follows you around you're golden. If all signals are go, and you guys make a quiet second of eye contact (as NitrousMafia said, you'll know) make your move.
